Project title: “Epigenetics of periodontitis: DNA methylation in disease development and as a potential therapeutic target.”

Project description:
The aim of this project is to analyze pathological changes in DNA methylation in gingival cells from patients with periodontitis, identify the influence of Porphyromonas gingialis on DNA methylation in these cells and characterize its biological consequences. Studies performed within this project will not only improve our understanding of this disease, but can also facilitate identification of new therapeutic targets, constituting a foundation for testing new drug classes in the treatment of periodontal disease in the future.  
 
Key responsibilities include:
  1. Analyses of effects of DNMT inhibitors on gingival fibroblast (GF) and periodontal ligament fibroblast (PLF) activation, proliferation and viability in the context of P. gingivalis infection. 
  2. Functional studies of antimicrobial responses of GFs and PLFs upon treatment with DNMT inhibitors (phagocytosis and bacterial elimination). 
Profile of candidates/requirements:
  1. BSc degree (or equivalent) in biotechnology, biochemistry or any related field of life sciences. 
  2. Strong experience in eukaryotic cell culture and in basic cell biology techniques (ELISA, Western Blot, qPCR) evidenced by results presented in BSc thesis or publication. 
  3. Experience in epigenetic studies and culturing anaerobic bacteria would be an advantage. 
  4. Fluency in written and spoken English. 
  5. Good skills in work organization, planning and keeping experimental track record. 
  6. High level of independence, determination and reliability, willingness to work outside of standard office hours. 
  7. Ability to work in a team.
